Инкубация in vivo гидрокортизон-резистентных тимоцитов в присутствии непептидного митогенного фактора приводит к накоплению экстра- и внутриклеточного гипоксантина, происходящего из 8-14C-адениловой кислоты. Стимуляция in vitro инкорпорации 2-14C-тимидина в ДНК тимоцитов может осуществляться с помощью соматотропного гормона в случае использования несепарированных тимоцитов (но не гидрокортизонрезистентных). Не наблюдалось накопления гипоксантина в соматотропин-стимулированных несепарированных тимоцитах крыс. Инкорпорацию 2-14С-тимидина в гидрокортизон-резистентную ДНК тимоцитов усиливали инкубацией клеток в присутствии гипоксантина в концентрациях, наблюдаемых после стимуляции тимоцитов непептидным митогенным фактором. Авторы приходят к выводу, что соматотропин и непептидный митогенный фактор стимулируют пролиферацию двух различных популяций тимоцитов, которые могут быть вовлечены в процесс пролиферации и регенерации тимуса под воздействием стероидов</p></abstract><trans-abstract xml:lang="en"><p>Hydrocortisone-resistant thymocyte proliferation is stimulated by the nonpeptide mitogenic factor. In vivo incubation of hydrocortisone-resistant thymocytes in the presence of the nonpeptide mitogenic factor results in accumulation of extra- and intracellular hypoxanthine, originating from 8-14C-adenylic acid. Stimulation of in vitro incorporation of 2-14C-thymidine in the thymocyte DNA may be effected via the somatotropic hormone in case nonseparated thymocytes (but not hydrocortisone-resistant ones) are used. No hypoxanthine accumulation in somatotropin-stimulated nonseparated rat thymocytes was observed. 2- 14C-thymidine incorporation in the hydrocortisone-resistant thymocyte DNA was enhanced by incubation of the cells in the presence of hypoxanthine in concentrations observed after thymocyte stimulation with nonpeptide mitogenic factor.
