полиморфного локуса Glu23Lys гена KCNJ11 с развитием сахарного диабета 2-го типа (СД2) в кыргызской популяции.</p><sec><title>Материал и методы</title><p>Материал и методы. Обследованы 287 человек кыргызской национальности. Исследование было проведено по типу «случай—контроль», включало 178 больных с СД2 (55 женщин и 123 мужчины, средний возраст 54±6,6 года) и 109 пациентов (48 женщин и 61 мужчина, средний возраст 50±8,4 года) без нарушений углеводного обмена (контрольная группа). Идентификация аллелей в полиморфном локусе Glu23Lys гена KCNJ11 проводилась методом регистрации полиморфизма длин рестрикционных фрагментов.</p></sec><sec><title>Результаты</title><p>Результаты. При сравнительном анализе распределения частот генотипов и аллелей полиморфизма Glu23Lys гена KCNJ11 среди больных СД2 и лиц контрольной группы выявлены статистически значимые различия. Частота встречаемости аллеля 23Lys в гетерозиготном и гомозиготном состояниях (Glu23Lys+Lys23Lys) у больных СД2 была значимо выше, чем в группе контроля (χ2=4,09; р=0,043). Риск развития СД2 у носителей аллеля 23Lys увеличивается в 1,46 раза [OR=1,46 (1,02—2,07); р=0,036]. Гомозиготный генотип Glu23Glu и аллель Glu23 снижают риск развития СД2 [OR=0,61 (0,37—0,99); p=0,043 для генотипа и OR=0,69 (0,48—0,98); p=0,037 для аллеля].</p></sec><sec><title>Заключение</title><p>Заключение. В кыргызской популяции полиморфный локус Glu23Lys гена KCNJ11 ассоциирован с развитием СД2. Предрасположенность к СД2 наследуется согласно доминантной и аддитивной моделям наследования признака. У кыргызов аллелем риска развития СД2 в полиморфном локусе Glu23Lys гена KCNJ11 является 23Lys.</p></sec></abstract><trans-abstract xml:lang="en"><p>Aim — in this study, we investigated whether polymorphisms Glu23Lys in KCNJ11 are associated with Type 2 diabetes mellitus (T2DM) in the Kyrgyz population.</p><sec><title>Material and methods</title><p>Material and methods. We genotyped 287 Kyrgyz individuals. A case—control study was performed, including 178 patients with T2DM (female — 55, male — 123, average age 54±6,6) and 109 non-diabetes controls (female — 48, male — 61, average age 50±8,4). The genotypes of polymorphic position Glu23Lys of KCNJ11 were determined by PCR-RFLP assay.</p></sec><sec><title>Results</title><p>Results. Genotypes Glu23Lys and Lys23Lys, containing the minor allele 23Lys, were more frequent in the group of type 2 diabetes (χ2=4,09; p=0,043). The allele 23Lys of KCNJ11 gene is associated with a high risk of developing type 2 diabetes in the Kyrgyz population [OR=1,46 (1,02—2,07); p=0,036]. Homozygous genotype Glu23Glu and allele Glu23 reduces the risk of developing type 2 diabetes [OR=0,61 (0,37—0,99); p=0,043 and OR=0,69 (0,48—0,98); p=0,037], respectively).</p></sec><sec><title>Conclusion</title><p>Conclusion. Polymorphisms Glu23Lys of KCNJ11 gene are associated with the risk of Type 2 diabetes mellitus in the Kyrgyz Population according to the dominant and additive models of inheritance of the trait.
